Description

PE-22-28 is a synthetic heptapeptide with the sequence Gly-Val-Ser-Trp-Gly-Leu-Arg, corresponding to residues 22-28 of the sortilin propeptide (UniProt Q99523; sortilin is also designated neurotensin receptor 3, NTSR3). It is the shortened analogue of spadin, which spans residues 12-28 of the same propeptide. Molecular formula C35H55N11O9, molecular weight 773.9 Da. Research applications focus on TREK-1 potassium channel modulation and neuronal signaling.

Research Summary

PE-22-28 is a relatively novel peptide under investigation for neurotrophic properties. Preclinical studies suggest it may promote neuronal survival and enhance synaptic plasticity through modulation of TREK-1 potassium channels. Research is in early stages, primarily limited to in vitro and rodent models.

Mechanism of Action

Proposed to inhibit TREK-1 two-pore domain potassium channels, leading to increased neuronal excitability and enhanced neurotransmitter release. This mechanism parallels the action of spadin, a natural TREK-1 inhibitor linked to neurotrophic signaling.
